Exemestane (also known as Aromasin) is a member of the class of compounds known as aromatase inhibitors. Aromatase is simply an enzyme that synthesizes estrogen. Aromatase inhibitors block the synthesis of estrogen. Exemestane is an irreversible, aromatase inactivator, structurally related to the natural substrate androstenedione. It acts as a false substrate for the aromatase enzyme, and is processed to an intermediate that binds irreversibly to the active site of the enzyme causing its inactivation

Of note: The estrogen suppression rate for Exemestane varies from 35% for estradiol (E2) to 70% for estrone (E1).

ESTROGEN SUPPRESSION METHOD
It restricts the levels of circulating estrogen, which is present in animal test subjects. The ovaries are the main source of estrogen, however, post-menopause, the source of estrogen is the product from the aromatization of androstenedione and testosterone. This reaction is facilitated by aromatase, which is a member of the cytochrome P-450 (CYP) family. Aromatase is found in the liver and adrenal glands, as well as in fat tissue.

Exemestane has an androstenedione-like structure. This means that it can compete with androstenedione and testosterone. It makes covalent bonds with the substrate-binding part of the enzyme. This action leads to eventually irreversible aromatase inactivation.
